ERP44 (endoplasmic reticulum protein 44) gene encodes a protein that contains a thioredoxin domain with a CRFS motif at the N-terminus and belongs to the protein disulphide isomerase (PDI) family of proteins. It is induced upon ER stress and forms mixed disulfides with substrate proteins through its conserved CRFS motif. It functions in thiol-mediated retention of proteins, such as Ero1α and Ero1β, which regulate oxidative protein folding in the ER. It also inhibits the Ca²⁺ channel activity of inositol 1,4,5-trisphosphate receptor type I.
