Methylene blue is bis(dimethylamino)phenothiazine hydrochloride (tetramethylthionine),
C₁₆H₁₈N₃SCl. It is a major histological dye in pharmacology and microbiology. Methylene blue is tested for histological and and bacteriological staining, and as a constituent of Wright′s blood stain.

Methylene blue is widely used in several routine staining applications including Mallory′s phloxine-methylene blue on paraffin sections of tissue fixed in Zenker′s fluid for nuclear staining and bacterial staining in milk. It is also suitable for manufacturing the polychrome methylene blue used in the preparation of Romanowsky stains.
